研究问题
Diabetic foot ulcers (DFU) remain a major clinical challenge due to their chronicity, high recurrence rates, and the risk of lower-limb amputation.
方法
After 24 h, the cells were pre-treated with the inhibitors for 30 min and then exposed to the compound VCE-005.1 for 6 h.;The inhibitors used included Dorsomorphin (DS) (AMPK inhibitor #S7306, Selleckchem, Houston, TX, USA), EX527 (SIRT1 inhibitor #E7034, Sigma-Aldrich, St.;Using a TriStar2 Berthold/LB942 multimode reader (Berthold Technologies, Bad Wildbad, Germany), luciferase activity was measured according to the instructions provided by the luciferase assay kit (Promega, Madison, WI, USA).;Western Blots] After stimulation and washing with phosphate-buffered saline (PBS), proteins were harvested using 50 μL of RIPA buffer containing phosphate and protease inhibitors (50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 2 mM EDTA, 1% NP40, 0.5% sodium deoxycholate, 0.5 mM Na3VO4, 50 mM NaF, and 40 μg/μL Protease Inhibitor Cocktail) or with lysis buffer previously described by [18].
核心结论
A significant Treatment × Day interaction was observed in db/+ mice (p = 0.046), whereas no significant interaction was detected in db/db mice.;A significant treatment × day interaction was observed in db/+ mice (p = 0.040), whereas the interaction did not reach statistical significance in db/db mice (p = 0.061). ‖ 结论:The present study demonstrates that VCE-005.1 promotes diabetic wound healing through a coordinated mechanism involving endothelial protection, vascular regeneration and resolution of inflammation.
局限
Although several pro-angiogenic strategies, including growth factor- and cell-based therapies, have shown encouraging preclinical results, their clinical translation has been limited by modest efficacy, poor persistence at the wound site and safety concerns [1,2,3,4,5,40].;Although the precise molecular hierarchy remains to be established, our findings support a coordinated signaling network that promotes endothelial adaptation to metabolic and hypoxic stress.;The present study has several limitations that should be considered in the context of further development.
