研究问题
The biological effects of SIRT1 during aging may therefore depend not only on its abundance or activity but also on whether the cell can engage, coordinate, and recover SIRT1-dependent responses as metabolic and redox demands change.
方法
Three concepts organize the framework used in this review.;In this review, the term “redox” is used broadly to include reactive oxygen species (ROS) and reactive nitrogen species (RNS), as well as NAD+/NADH, NADP+/NADPH, GSH/GSSG, and thioredoxin systems.;The adaptive landscape proposed in this review does not represent a fixed sequence of molecular states.;Consistent with this, a 2026 systematic review of 33 human intervention studies found relatively consistent increases in circulating or cellular NAD-related metabolites after oral NR or NMN, but inconsistent or often null effects on functional, metabolic, and vascular outcomes [208].
核心结论
Cellular senescence is itself heterogeneous: p16 and p21 expression, SA-β-gal activity, and SASP components vary across cell types, tissues, and senescence-inducing conditions, and no single marker is sufficient to define senescence across biological contexts [95,96,113,114].;For example, SIRT1 depletion in human umbilical vein endothelial cells induces a premature senescence-like phenotype with increased SA-β-gal activity and sustained growth arrest [116,117].;In human neural stem cells, amyloid-β-induced senescence-like changes are associated with reduced SIRT1, PGC-1α, and FOXO3 expression, whereas SIRT1 overexpression partially attenuates these alterations [130]. ‖ 结论:Testing these relationships experimentally will be important for determining when modulation of SIRT1 can contribute to restoration of cellular adaptation and when additional components of the aging cellular environment must also be addressed.
