研究问题
Cancer is a leading cause of death worldwide, driving continued research for innovative therapies with enhanced specificity and efficacy.
方法
Natural products have long been recognised as a valuable source of anticancer agents due to their structural diversity and unique biological activities.
核心结论
Building on the anticancer potential of oleuropein, a major olive tree (Olea europaea) metabolite, we evaluated a newly synthesised semi-synthetic analogue, GS32, for its cytotoxic activity in vitro.;Cytotoxicity assays revealed that GS32 exhibited broad-spectrum activity against various cancer cell lines, while also displaying differential toxicity between normal and cancerous cells.;Through transcriptomic analysis, we discovered that GS32 differentially disrupts key cellular pathways involved in stress response.;Importantly, targeted phosphoproteomic profiling uncovered context-dependent modulation of key signalling pathways, including the AKT/mTOR and NF-κB axes, as well as stress-response regulators. ‖ 结论:This study provides new mechanistic insight into the cellular effects of GS32 and identifies the compound as a candidate that merits further preclinical investigation.
