研究问题
Parkinson’s disease (PD) is a progressive neurodegenerative disorder characterized by the selective loss of substantia nigra dopaminergic neurons and subsequent striatal dopamine depletion, resulting in cardinal motor symptoms including resting tremor, rigidity, bradykinesia, and postural instability.
方法
Particle Size, Polydispersity Ind] The colloidal characteristics of the Mexil-loaded niosomal dispersions were measured as mean hydrodynamic diameter, polydispersity index (PDI), and zeta potential using Zetasizer Nano ZS (Malvern Instruments, Malvern Instruments Ltd., Malvern, Worcestershire, UK) at 25 °C.;Each formulation was measured in three replicates, and the results are represented as mean ± standard deviation (SD; n = 3) [78].;Samples were evaluated at 1, 3, and 6 months for PZ, ZP, PDI, and the percentage of entrapped Mexil in the niosomal formulation.;Animals] In a controlled environment with a constant temperature of 23 ± 2 °C, a 12 h light/dark cycle, and a humidity of 50 ± 10%, adult male Sprague–Dawley rats weighing an average of 200–250 g were maintained (4 rats/cage).
核心结论
Formulations prepared with higher lecithin content (225 mg; N4–N6) were found to have significantly higher EE% (84.2–93.0%) than those with 150 mg of lecithin (58.0–71.0%).;Among all of the formulations, N4 showed the highest EE% (93.0 ± 1.7%), followed by N6 (89.1 ± 2.6%) and N5 (84.2 ± 1.5%). ‖ 结论:Overall, our results suggest that niosomal Mexil exerts robust neuroprotective effects against rotenone-induced Parkinsonism-like neurodegeneration by acting on the co-regulation of oxidative stress, inflammation, mitochondrial dysfunction, autophagy, and apoptosis.
