研究问题
Alzheimer’s disease (AD) is a complex neurodegenerative disorder in which amyloid-β (Aβ) deposition, abnormal tau phosphorylation, neuroinflammation, synaptic impairment, and progressive neuronal loss are major pathological features [1].
方法
Importantly, direct evidence that any of these five minor phytocannabinoids modulate ER stress in AD-relevant neuronal models is currently lacking, and most mechanistic links discussed in this review are derived from non-neuronal systems or non-AD disease models.;As this article is a narrative mechanistic review rather than a systematic review, relevant literature was identified primarily through PubMed searches using combinations of terms related to Alzheimer’s disease, endoplasmic reticulum stress, unfolded protein response, proteostasis, and the five minor phytocannabinoids discussed in this review.;In this review, such overlap is therefore used as a hypothesis-generating framework for identifying testable compound–pathway relationships rather than as evidence of established mechanistic relevance or therapeutic benefit.;Some studies cited in this review employed cannabinoid-enriched extracts rather than isolated compounds [57,58].
核心结论
Introduction Alzheimer’s disease (AD) is a complex neurodegenerative disorder in which amyloid-β (Aβ) deposition, abnormal tau phosphorylation, neuroinflammation, synaptic impairment, and progressive neuronal loss are major pathological features [1].;Disruption of XBP1s nuclear translocation, impaired interaction with phosphoinositide 3-kinase (PI3K) regulatory subunits p85α/β, or SNO-mediated suppression of IRE1α ribonuclease activity may weaken the adaptive IRE1α–XBP1s pathway and contribute to unresolved protein misfolding [32,33,39,40].;In THC-dominant cannabis flowers, tetrahydrocannabinolic acid or total THC commonly occurs at percent-level concentrations, often in the range of approximately 10 to more than 20% by dry weight in commercial or medical cannabis datasets [54,55]. ‖ 结论:Therefore, minor phytocannabinoids may serve as pharmacological probes for testing whether the upstream modulation of these processes can support ER proteostasis in AD-relevant systems.
