研究问题
Glutamate is the primary excitatory neurotransmitter in the central nervous system (CNS) that mediates excitatory synaptic transmission [1].
方法
Eight-week-old male C57BL/6 mice, weighing approximately 25–30 g, were employed in the in vivo experiments.;Animals were housed 2–3 per cage in the animal facility of the Institute of Molecular Biology and Biotechnology (IMBB)—Foundation for Research and Technology Hellas (FORTH) under stable conditions in a 12 h light–dark cycle and with food and water provided ad libitum.;All experimental procedures were performed by skilled individuals, holders of FELASA certificate, and were conducted, taking into consideration animal well-being and reduction in suffering, in accordance with the 3R principles, the EU directives (2010/63/EU and L 276/33/20.10.2010) and the Greek legislation (Presidential Decree 56/30.04.2013) for the care and use of laboratory animals.;For in vivo experiments, AM11920 was further diluted in NaCl 0.9% at a concentration of 0.5 mg/kg.
核心结论
结论:Collectively, the present study demonstrates that pharmacological enhancement of endogenous 2-AG signaling through inhibition of its primary hydrolyzing enzymes, MAGL and ABHD6, by AM11920, confers significant neuroprotection and exerts broad immunomodulatory control over excitotoxic inflammation induced by AMPA.
局限
Thus, the extent to which CB2R mediated the neuroprotective and anti-inflammatory actions of AM11920 remains to be established.;Another limitation of the present study is the dose of AMPA employed for the in vitro experiments (150 μΜ), which represents a robust excitotoxic challenge and may not directly reflect the concentration of glutamate, observed in vivo under excitotoxic conditions.;Apart from this, the lack of behavioral assessment represents another limitation of this study and therefore it remains unclear whether the protection observed at the cellular level translates into functional improvement too.
