研究问题
Alzheimer’s disease (AD) is a progressive neurodegenerative disorder considered to be one of the most common causes of cognitive decline.
方法
In addition, because global redocking of KYT-36 did not reproduce the crystallographic pose, a local optimization starting from the experimental KYT-36 geometry was performed to assess compatibility of the prepared Kgp receptor with the crystallographic binding mode.;Interaction Guided Dual-Target Prio] Pose refinement included analysis of hydrogen bonds, hydrophobic contacts, π-stacking, π-cation interactions, salt bridges, and halogen bonds, as well as other PLIP-supported interactions through the protein–ligand interaction profiler [24].;These included predicted blood–brain barrier permeation, confirmation that it is not a P-glycoprotein substrate, a molecular weight of 357.41, a TPSA of 54.79 Å2, and, crucially, no PAINS or Brenk alerts.;The calculated energetic components included van der Waals, electrostatic, polar solvation, and nonpolar solvation terms.
核心结论
Protein–ligand heteroatom distances of ≤3.5 Å were observed in 8.6% of analyzed frames, while distances of ≤4.0 Å occurred in 57.0% of frames, indicating intermittent polar proximity rather than a sustained hydrogen-bond interaction. ‖ 结论:Therefore, Kgp should only be considered as an investigative target rather than a clinically validated AD target.
局限
These observations are preliminary because independent MD replicates were not performed.
对本项目的价值
★★☆☆☆ 脑老化是最大未满足需求,也是终点最难量化的领域;biomarker 与认知读数选择最敏感。
