研究问题
Whether this is just due to the increase in concentration of the ‘active compound’ and/or is influenced by the presentation of the peptide motif at high density on the surface of the micelles, remains an open question. The Picrosirius red assays, both quantitative by absorbance assays and qualitative by polarized microscopy, indicates a significant increase in collagen when compared to controls, showing that FMdEG-KTTKS has a substantial collagen-stimulating effect, at low doses (lowest concentration 0.00625 wt% examined) which is beneficial for potential applications in skincare.
方法
The KTTKS pentapeptide is based on a sequence taken from a propeptide present in the Human type I collagen sequence.3,4 It has been reported that both the peptide KTTKS3 and C16-KTTKS5 are able to promote type I collagen biosynthesis in vitro.;The distinct N-terminal moiety compared to simple unbranched lipids leads to self-assembly into spherical micelles, contrasting with β-sheet nanotape formation of the C16-KTTKS or C18-KTTKS10 lipopeptides (C14-KTTKS showed presented an unordered conformation10).;We also show that FMdEG-KTTKS performs as an anti-inflammatory molecule, reducing nitrite production by macrophages after exposure to bacterial lipopolysaccharide.
核心结论
Studies also suggest that C16-KTTKS can reduce the appearance of facial wrinkles and may help in the recovery of skin wounds.6–8 Although it has been established that KTTKS improves the production of type I collagen in vitro, an important consideration in the topical application of peptide amphiphiles is how to improve the transport of the bioactive peptide across the stratum corneum, a natural barrier against exogenous compounds, and the lipidated conjugate was developed to improve delivery across the epidermis.;Together, these findings position FMdEG-KTTKS as a versatile candidate for potential future applications in cosmetics due to collagen-stimulating properties, and in regenerative medicine due as it promotes wound healing.ExperimentalMaterialsConjugate Farnesyl-mercaptopropyl-(EG)2-KTTKS (EG: ethylene glycol), denoted FMdEG-KTTKS, was synthesized using automated solid phase Fmoc peptide synthesis by Peptide Protein Research Ltd (UK).;The anti-inflammatory effect may be attributed to the influence of the farnesyl group in FMdEG-KTTKS, for which anti-inflammatory effects have been noted previously, including regulation of the response to oxidative stress and controlling cell migration.62–64 Our findings suggest that FMdEG-KTTKS may not only aid with collagen production and cell repopulation after injury, but also act as a potential anti-inflammatory molecule.ConclusionsWe present a comprehensive biophysical and biological characterization of a novel matrikine lipopeptide, FMdEG-KTTKS.
局限
For these higher concentrations however, the high q form factor oscillations are retained, indicating that the internal micelle structure is little changed (as also shown by the fit parameters, except τ which represents an overall scale factor of scattering contrast, dependent on concentration).Fig. 1Maximum intensity, I0, and position, λ0, of the fluorescence emission peak of solutions containing ANS, plotted as a function of the FMdEG-KTTKS concentration.Fig. 2Conformation and self-assembly data.
