研究问题
In the present study, we investigated (i) the role of gut microbes in the metabolism of blueberry phytochemicals using germ-free mice and antibiotic-treated mice, (ii) the impact of depleting microbes on the vascular benefits of dietary blueberries, and (iii) the association between gut microbiome, blueberry metabolome, and vascular inflammation.2.Materials and methods2.1.Experimental designSix-week-old male C57BL/6J mice (Stock Number: 000664) were purchased from the Jackson Laboratory (Bar Harbor, ME, USA).;CB for relative abundance of taxa.3.5.Gut microbiota depletion reduces blueberry-derived metabolites and abolishes the benefits of dietary blueberries on vascular inflammationCohort 3 aims to determine whether the benefits of dietary blueberries on vascular inflammation are mediated through microbiome–metabolome interaction.;We assessed whether dietary blueberries affect metabolic parameters to determine whether the reduced vascular inflammation with blueberries are due to a secondary effect.

核心结论
Additionally, 3,4-dimethoxycinnamic acid increased 60-fold in the CB group, but this increase was reduced by antibiotic treatment. p-Coumaric acid is present in many fruits and vegetables, but is not present in the blueberry powder used in our present study.27 Our findings suggest that p-coumaric acid may be produced by host enzymes, while hippuric acid and 3,4-dimethoxycinnamic acid could be generated by gut microbial enzymes.;Collectively, the genus enriched with blueberry supplementation is positively associated with blueberry-derived metabolites, whereas the genus reduced with blueberry supplementation is negatively associated with blueberry-derived metabolites.Fig. 2(A) Blueberry-derived plasma metabolites in the standard diet condition.;Metabolite analysis indicates that HFD decreases the levels of many circulating metabolites compared to mice fed a control diet (HF versus C mice) (Fig. 3B).
局限
However, Extibacter, which is reduced with blueberry supplementation, is negatively associated with hippuric acid and 3,4-dimethoxy cinnamic acid.;However, antibiotic treatment abolished the anti-inflammatory benefits conferred by dietary blueberries, and no significant differences in vascular endpoints were observed in HBA vs.;However, supplementation with blueberries and/or antibiotic treatment did not alter these metabolic parameters (Table S3).Fig. 3(A) Indices of vascular inflammation.
