研究问题
We hypothesized that the two animal species would exhibit distinct high‐altitude adaptation strategies, specifically reflected in the rumen system, as follows: (1) differences in the structure and function of the rumen microbiome to mediate host energy supply, influencing high‐altitude adaptability, and (2) in response to differences in rumen‐derived energy supply, the absorption and metabolic functions of the rumen epithelial cells differ.;Subsequently, we transplanted the rumen microbiome of yaks into Holstein cows to verify whether the transfer could improve the host's adaptation and performance.;In this respect, 6 h after transplantation was considered to represent the immediate response phase, allowing the rumen microbiome of present and transplanted rumen fluid to settle, 7 days after transplantation to reflect early colonization dynamics, and 28 days after transplantation to afford early indications of a likely equilibrium establishing and to illustrate whether or not the transplanted rumen microbiome persisted.
方法
We selected Holstein cows for this purpose because this breed is of pure lowland origin and has been bred for extremely high performance.;We employed a multi‐omics approach, including metagenomics and meta‐transcriptomics; further, we applied single‐nuclei RNA sequencing (snRNA‐seq) to rumen epithelial cells as an indirect means of assessing cell‐type‐specific functions.;We hypothesized that the two animal species would exhibit distinct high‐altitude adaptation strategies, specifically reflected in the rumen system, as follows: (1) differences in the structure and function of the rumen microbiome to mediate host energy supply, influencing high‐altitude adaptability, and (2) in response to differences in rumen‐derived energy supply, the absorption and metabolic functions of the rumen epithelial cells differ.
核心结论
However, these findings may be confounded by dietary and altitudinal factors. This study aimed to systematically investigate the differences in high‐altitude adaptation strategies between indigenous yaks and a closely related species, cattle (Bos taurus), from the perspective of rumen ecology.;Based on these findings, we conducted a Wilcoxon rank‐sum test at the genus level to analyze the differential abundance of the top five shared bacterial genera between the two groups (Figure 1E).;Based on these findings, we focused on the expression of genes involved in energy‐metabolism‐related pathways, including volatile fatty acid transport and ketone‐body metabolism.
局限
However, research on the differences in the gastrointestinal microbiome and their role in the environmental adaptation of yaks is lacking.;However, these findings may be confounded by dietary and altitudinal factors. This study aimed to systematically investigate the differences in high‐altitude adaptation strategies between indigenous yaks and a closely related species, cattle (Bos taurus), from the perspective of rumen ecology.;However, to the best of our knowledge, no detailed description of this mechanism is currently available.
