研究问题

In the pre- sent study, we aimed to determine whether chronic alcohol administration alters AhR- and Nrf2-related signaling and whether sesaminol supplementation is associated with an amelioration of any effect.;Gut tissue sections (4-μm thick) were prepared and stained with periodic acid-Schiff (PAS) to evaluate mucosal architecture, and liver sec- tions were similarly processed and stained with Oil Red O to visualize lipid accumulation.;Protein analysis and quantification Western blotting To evaluate colonic protein expression, 100 mg of each tissue sample was lysed in 1 mL of M-PER Mammalian Protein Extraction Reagent (Thermo Fisher Scientific, Waltham, MA, USA) containing a protease inhibitor cocktail (Nacalai Tesque).

方法

Graphical Abstract Keywords Cellular senescence · Alcohol · Sesaminol · Gut microbiota · Aryl hydrocarbon receptor · Nuclear factor erythroid 2-related factor 2 Introduction Data collected globally from diverse cohorts indicate that alcohol use is increasingly prevalent among older adults [1].;In our previous studies of young mice, we established that chronic alcohol intake triggers oxidative stress and inflammation in the gut, resulting in chronic colitis, gut leakiness, and dysbiosis, consistent with a “multi- hit” mechanism for the pathogenesis of alcohol-related colorectal cancer (ARCRC) [10–12].;Experimental groups and housing conditions Young (8 weeks old, n=8) and middle-aged (48 weeks old, n=32) male C57BL/6NCr mice were purchased from SLC (Hamamatsu, Japan).

核心结论

Taken together, these findings suggest that sesaminol may ameliorate the interconnected microbial, metabolic, epithelial, and redox-related responses to chronic alco- hol exposure, thereby attenuating alcohol-associated intestinal dysfunction and senescence-related changes in middle-aged mice.;Furthermore, we demonstrated that oral sesa- minol supplementation ameliorates these patho- physiological changes [11, 12].;These findings indicate that sesaminol supplementation was associated with increased fecal butyrate levels.

局限

However, Group A-ES was character- ized by a restoration of Nrf2 activity to a level com- parable to that of Group A-C (Fig.;However, chronic alcohol administration (Group A-E) resulted in a downward trend in lactate concentrations (Fig.
