研究问题
Alzheimer's disease (AD) is characterized by the accumulation of amyloid beta (Aβ) and tau pathology, with tau pathology being more closely linked to the emergence of dementia symptoms.
方法
20 The sample comprised 202 cognitively unimpaired (CU) participants, 162 participants with mild cognitive impairment (MCI), and 77 participants with AD dementia.;Interpretation: Our findings demonstrate that genetic risk variants, which cluster primarily on genes involved in vesicular trafficking and endosomal transport, modulate amyloid‐related regional fibrillar tau: individuals with higher transport polygenic risk scores showed higher levels and higher rates of tau positron emission tomography increases during amyloid accumulation as measured by time since becoming first amyloid positive.
核心结论
Regression lines and 95% CI (shaded areas) are presented for binarized groups defined by pPRS < 25th percentile (blue, “Low pPRS”) and > 75th percentile (red, “High pPRS”). ‖ 结论:We provide genetic evidence, most robust in a well‐powered discovery cohort and directionally consistent in an independent cohort, implicating intracellular transport pathways in tau accumulation in AD.
局限
A further limitation concerns the granularity of our pathway definitions.;Resolving pathway clusters into narrower, adequately powered subcomponents is a direction for future work in larger cohorts.;A4's exclusively CU composition and comparable‐to‐higher amyloid PET burden relative to ADNI further raises the possibility that cognitive resilience or unmeasured factors such as vascular burden and co‐pathology differ between cohorts and differentially influence the amyloid–tau relationship, a residual contribution of such unmeasured factors cannot be fully excluded, 65 and future work incorporating vascular imaging and co‐pathology biomarkers would help clarify this.
