研究问题

Standard antiepileptic drugs (AEDs), including phenobarbital and potassium bromide, remain the cornerstone of treatment; however, they fail to achieve adequate seizure control in 20%–30% of patients (; ; ).;However, despite promising preclinical evidence, few clinical studies have evaluated the efficacy of curcuminoids in naturally occurring canine epilepsy.;Moreover, the molecular mechanisms underlying their potential therapeutic effects remain unknown.

方法

The study protocol was approved by the Mahidol University Institute Animal Care and Use Committee of the Faculty of Veterinary Science (Protocol No.;Eight client-owned dogs with PES were enrolled alongside five healthy adult dogs (aged 1–6 years; mixed breeds and purebreds; no history of neurological disease, seizures, or systemic illness) recruited from the same institution as a proteomic reference control group.;Multi-layered statistical filtering (ANOVA, FDR correction, and VIP scoring) was applied to minimize false discoveries in the high-dimensional dataset.;Validation in larger, prospectively powered cohorts is required.

核心结论

Clinical response (≥25% reduction in seizure frequency and/or ≥1 grade reduction in seizure severity) was observed in four dogs (50%) ().;Within responder group, three dogs (No. 1, 2, and 6) exhibited decreased seizure frequency, and one dog (No. 7) showed improved seizure symptoms with reduced clonic movements.;In the four non-responder dogs (No. 3,4, 5, and 8), seizure frequency and severity remained stable compared with baseline, with no significant changes being observed. ‖ 结论:These findings support the integration of curcuminoids into multimodal epilepsy management strategies in veterinary medicine, with potential translational implications for human epilepsy therapy.

局限

In this uncontrolled pilot study (n = 8), curcuminoid supplementation was safe, well tolerated, and yielded a clinical response rate of 50% in dogs with PES; however, the absence of a concurrent control group limits interpretation to preliminary evidence of an association rather than definitive evidence of efficacy.;The 50% clinical response rate observed in this preliminary is comparable to those reported for second-line AEDs in canine epilepsy ().;The finding that clinical improvement occurred in only 50% of dogs suggests potential heterogeneity in therapeutic response, which may be related to individual differences in curcuminoid bioavailability, genetic polymorphisms affecting drug metab
