研究问题

Dementia, a syndrome marked by impairments in memory, reasoning, and judgment, severely impacts daily functioning and has a significant emotional and economic burden.

方法

This specialized clinic, held every Wednesday in the Geriatric Medicine department of a tertiary care center, focuses on the comprehensive evaluation of patients referred for cognitive decline assessment.;The study was approved by the institutional ethics committee (IEC PG-567/26.09.19), and all participants were provided with comprehensive information about the study and informed consent was obtained from all participants before their inclusion in the study.;For patients with AD, informed consent was obtained from a legally authorized representative (LAR).;[Data collection and procedure] Data collection involved sequential screening, clinical evaluation, and neuropsychological testing of participants, followed by venous sampling for serum NfL estimation.

核心结论

The participants were predominantly male 72.1%. ‖ 结论:These findings support the potential of serum NfL as a non-invasive biomarker for early detection and staging of cognitive decline.

局限

Future studies with balanced gender representation are needed to clarify potential sex-related differences.;This underscores the importance of addressing gender disparities in future research to ensure generalizability [23–25].;However, the proposed ROC-derived cut-offs are preliminary, likely cohort-specific, and should be interpreted as exploratory rather than definitive diagnostic thresholds.

对本项目的价值

★★★☆☆ 脑老化是最大未满足需求,也是终点最难量化的领域;biomarker 与认知读数选择最敏感。最大样本量约 129;设计 Meta 分析、系统综述。
