研究问题

Assessment of short-term within-subject variability and test-retest reliability of Alzheimer's disease (AD)-related plasma biomarkers is important for real-world clinical implementation and interpretation in research studies.

方法

Participants from research studies at the Demen- tia Research Centre, University College London (UCL), which recruit individuals with and without cognitive impairment, were included if they had at least 2 serial blood sample collections, as part of their study enrol- ment, taken a maximum of 3 months apart.;Alzheimer's Research & Therapy (2026) 18:238 Sample collection and processing Serial blood sampling was performed on 92 participants either on the same day or within 92 days (~3 months).;Participants were not asked to fast prior to sample col- lection.

核心结论

Each point represents an individual biomarker effect estimate (beta) and error bars represent the corresponding 95% confidence intervals (CIs).;Adjusted p-values are NS unless other indicated; *p<0.05, p<0.01, *p<0.001 Page 9 of 12 Beament et al.;Cullen et al. reported test-retest variability of 20.0% for p-tau217, here we report a CVw of 11.2% [33]. ‖ 结论:In conclusion, core AD biomarkers show strong correla- tions and high test-retest reliability, with low within-sub- ject variability across assay platforms.

局限

Although we limited inclusion to samples collected within three months, during which substantial neuro- degenerative changes are unlikely to occur, we observed increased variability over longer sampling intervals for some biomarkers, which may reflect underlying physi- ological changes and warrants further investigation.

对本项目的价值

★★☆☆☆ 脑老化是最大未满足需求,也是终点最难量化的领域;biomarker 与认知读数选择最敏感。最大样本量约 92;设计 Meta 分析、系统综述。
