研究问题
Neurodegenerative diseases (NDs) are a heterogeneous group of complex multifactorial disorders affecting millions of people worldwide [1].
方法
We searched publicly available scientific literature repositories, focusing on review articles published over the last 10 years and original research articles published primarily within the last 5 years.;Notably, a systematic review encompassing forty studies of urinary metabolomics in PD highlighted the potential of this approach to capture disease-associated metabolic alterations [105].;Of all the models discussed in this review, large mammals offer the highest overall translational potential across the three dimensions described above.;In this study, characterization of founder and F1knock-in pigs revealed consistent movement and behavioral abnormalities, and germline-transmissible early death.
核心结论
This combined approach revealed that extracellular amyloid-β (Aβ) plaques induce highly localized pre-synaptic toxicity and dystrophic neurites strictly restricted to the immediate plaque microenvironment, rather than causing widespread degeneration.;Furthermore, neuroscience research systematically underrepresents female donors, with female-derived lines comprising less than 30% of ND modeling cohorts.;By applying single-cell multi-omics to complex assembloids, researchers can detect non-cell-autonomous alterations within a healthy recipient organoid, driven exclusively by its physical coupling to a genetically altered or patient-derived donor culture [91]. ‖ 结论:In this context, a particularly promising direction for NDs research lies in strengthening the bidirectional connection between computational modeling and bedside clinical practice, ultimately facilitating the translation of mechanistic knowledge into more individualized therapeutic strategies.
