研究问题
Pain is an increasingly recognized, albeit heterogeneous, manifestation of neurodegenerative disorders.
方法
This systematic review evaluated preclinical evidence on the contribution of microglial and astrocytic alterations to pain and nociceptive modulation in animal models of neurodegenerative diseases.;Eligibility Criteria] Eligibility criteria were defined a priori according to the registered protocol and were structured around the animal population, neurodegenerative disease model, glial exposure or intervention, and pain-related outcomes.;Studies were considered eligible if they met all of the following criteria: (1) original, peer-reviewed experimental research using an in vivo animal model; (2) investigation of a neurodegenerative disease or an established experimental model reproducing relevant neurodegenerative pathology, including Parkinson’s disease, Alzheimer’s disease, multiple sclerosis/experimental autoimmune encephalomyelitis, amyotrophic lateral sclerosis, Huntington’s disease (HD), or another clearly defined neurodegenerative disorder; (3) assessment of microglial and/or astrocytic alterations, including cellular reactivity, morphology, glial marker expression,…
核心结论
The evidence base remained markedly uneven: sixteen studies (61.5%) investigated MS/EAE, seven (26.9%) investigated PD, two (7.7%) addressed AD-related models, and one (3.8%) investigated HD.;Thus, 23 of 26 studies (88.5%) were derived from MS/EAE or PD models, which limits cross-disease generalization.;Multiple Sclerosis and Experimental] MS/EAE represented the largest disease-specific evidence group, accounting for 16 of 26 included studies (61.5%). ‖ 结论:Collectively, these findings suggest that microglia and astrocytes should be regarded as dynamic components of neuroglial networks rather than as uniform markers or drivers of neuroinflammation.
局限
Convergence among IL-6/IL-6R, HMGB1, NF-κB, NLRP3, and inflammatory cytokine signaling supports a substantial neuroimmune component, but heterogeneity across the included studies indicates these mechanisms operate within broader neuroglial networks.;This heterogeneity is not merely a limitation of the available literature.;The limited incorporation of sex as an experimental variable in the included studies therefore represents both a methodological limitation and an important direction for future research.
