研究问题
Furthermore, it may be utilized to improve prognostic evaluation in AD by predicting disease progression and recovery potential via metabolic signatures, thus providing a supplementary objective reference for clinical decision-making.
方法
Furthermore, the underlying mechanisms by which metabolites play vital roles in AD pathology have also been elucidated [14–17].Here, we review recent advances in this field by summarizing metabolomics studies conducted on various types of clinical samples across different stages of AD.;We discuss the correlations between metabolites and AD pathologies, as well as their associations with cognitive function, along with the potential of metabolites to enhance the clinical diagnosis of AD.;We subsequently present an overview of the molecular pathways through which metabolites contribute to AD hallmark pathologies, which may provide insights for the development of targeted metabolic treatment strategies for AD.
核心结论
Recently, lipidomic and metabolomic studies have consistently demonstrated that diverse classes of lipids are altered or associated with AD neuropathology and cognition in AD brains (Table 1).;Metabolites that play a pivotal role in lipid metabolism, such as acylcarnitines, which are essential for mitochondrial β-oxidation of FAs [50], also demonstrated a correlation with AD traits [31].;However, multivariate analysis based on metabolites demonstrated a clear separation between AD, other dementias, and HC for serum samples, but not for saliva samples, indicating that serum might be more efficient and suitable for AD diagnosis than saliva [110].Gut microbial metabolites involved in ADIt is estimated that trillions of gut microbes exist in the human body, which undergo dynamic changes in response to physiological conditions, encode millions of genes, and produce a wide variety of metabolites [111, 112].
局限
It was not clear, however, what happens inside the brain as AD progresses in terms of metabolites [19].;However, multivariate analysis based on metabolites demonstrated a clear separation between AD, other dementias, and HC for serum samples, but not for saliva samples, indicating that serum might be more efficient and suitable for AD diagnosis than saliva [110].Gut microbial metabolites involved in ADIt is estimated that trillions of gut microbes exist in the human body, which undergo dynamic changes in response to physiological conditions, encode millions of genes, and produce a wide variety of metabolites [111, 112].;To address the above‑mentioned limitations, longitudinal and dynamic metabolic profiling covering preclinical and prodromal AD stages is urgently needed to identify early metabolic signatures and clarify the temporal causal relationship between metabolic dysregulation and AD pathological progression.
