研究问题
However, in recent years, companion animal biobanking has gained increasing attention, and multiple canine brain banks have now been established in Europe and North America (; ; ; ).;However, to the best of our knowledge, the extent to which lipofuscin accumulation in the canine brain is quantitatively associated with canine cognitive dysfunction in larger cohorts, as opposed to simply correlating with chronological age, is an open question.
方法
In early experiments, we performed cannulation of the carotid arteries.;The CT scanner we used was the OmniTom® Elite (NeuroLogica, Danvers, MA), a 16-slice scanner.;This was followed by lead aspartate staining at 60 °C.
核心结论
Of the 55 donors, 27 (49.09 %) had no reported signs of confusion, 27 (49.09 %) had signs ranging from mild to severe, and 1 (1.82 %) had unknown or unreported status.;The gross examination and CT scan modalities were significantly correlated (Spearman rho = 0.66, p = 0.0009, n = 22).;The histological vessel-clearance score was also positively correlated with both the gross score (rho = 0.76, p = 0.001, n = 15) and the CT score (rho = 0.70, p = 0.0017, n = 17), although these comparisons were limited by the smaller number of donors with histological grading. ‖ 结论:McKenzie are employees of Sparks Brain Preservation, a non-profit brain preservation organization.
局限
In our preliminary histologic investigations, we found that hippocampal lipofuscin burden is elevated in dogs with cognitive dysfunction.;However, we note that this histological analysis of lipofuscin is preliminary and exploratory.;We have also presented preliminary analyses of brain aging and ultrastructure that are intended to demonstrate its future research value.
