研究问题
Aim The aim of this narrative review is to explore and synthesize current knowledge on the adaptive responses of breast cancer cells to oxygen stress, with a particular focus on the mechanisms by which these cells survive and proliferate in hypoxic tumor microenvironments.;These new blood vessels aim to re-establish oxygen and nutrient delivery to the hypoxic tumor zones.;The balance between adaptive and terminal UPR signaling is therefore a critical determinant of whether oxygen-stressed breast cancer cells survive.
方法
Literature search strategy A comprehensive literature search was conducted across major biomedical databases, including PubMed, Scopus, Web of Science, and Google Scholar.;Methodological considerations No quantitative meta-analysis was performed due to variability in experimental models, endpoints, and clinical contexts.
核心结论
Through coordinated activation of hypoxia-inducible signaling pathways, breast cancer cells enhance angiogenesis, rewire metabolism toward glycolysis, suppress immune surveillance, and acquire invasive and stem-like phenotypes that collectively promote tumor progression and metastasis.
局限
However, under hypoxia, cancer cells shift toward glycolysis, a less efficient but oxygen-independent pathway, to sustain their energy needs.;However, the resulting vasculature is often structurally and functionally abnormal, leading to inefficient perfusion and fluctuating oxygen levels, thereby perpetuating a cycle of hypoxia and further tumor adaptation ( 11 , 12 ).
