研究问题
In Alzheimer’s disease (AD), the brain degenerates slowly over time.
方法
Animals] All methodologies involving animals in this study received the requisite approval from the Animal Ethics and Welfare Committee of Ningbo University.;The mice were sourced from Hangzhou Ziyuan Laboratory Animal Co., Ltd.;Twelve-month-old male wild-type (WT) and APP/PS1 mice (n = 10 per group) were maintained in a standardized, specific-pathogen-free environment at the Ningbo University Laboratory Animal Center under controlled environmental conditions.;The animals were provided with unrestricted access to food and water throughout the 7-day adaptation phase.
核心结论
Cells were retained only when mitochondrial gene expression was below 5%, and filtering thresholds of 200–5000 for nFeature_RNA and 1000–20,000 for nCount_RNA were applied. ‖ 结论:This study integrated transcriptomic and scRNA-seq data from patients with AD and revealed that ACO2 was significantly downregulated in the hippocampal tissues of patients with AD.
局限
The cross-sectional design also precluded the assessment of dynamic changes in ACO2 expression during AD progression, and all findings obtained from human samples should, therefore, be interpreted as associative rather than causal.;Animal experiments were also conducted using single-sex mice, which may limit the generalizability of the findings across sexes.;The relatively small clinical cohort may limit the generalizability and statistical robustness of our findings.
