研究问题
Diabetes Mellitus (DM), characterized by high blood sugar levels, is a serious chronic metabolic disease that is also frequently accompanied by hyperglycemia and various complications (including cardiovascular and chronic kidney diseases). , DM is becoming increasingly prevalent, according to the 2025 International Diabetes Federation Diabetes Atlas, approximately 589 million adults aged 20–79 years were living with diabetes worldwide in 2024, and this number is projected to increase to 853 million by 2050.;Among the two types of this metabolic disease defined as type 1 diabetes (T1DM) and type 2 diabetes (T2DM), T2DM is the most prevalent, and patients often suffer from wounds on the skin, most commonly on the lower extremities (especially the feet).
方法
The effects of the microneedle samples on modeled diabetic environment cell viability were evaluated using an indirect MTT assay.;For the viability assay, cells were seeded into 96-well plates at a density of 1 × 104 cells per well and incubated overnight to allow cell attachment.;The antioxidant and oxidant levels in the supernatants obtained from stressed HaCaT cells were determined using the Elabscience Total Antioxidant Status (TAS) Colorimetric Assay Kit (E-BC-K801-M) and the Elabscience Total Oxidant Status (TOS) Colorimetric Assay Kit (E-BC-K802-M), respectively, in accordance with the manufacturer’s instructions.;The migratory capacity of HaCaT human keratinocytes under simulated hyperglycemic conditions was assessed using a scratch wound gap closure assay.
核心结论
In contrast the characteristic lysine methylene resonance around 2.9 ppm, present in gelatin, was markedly reduced in GelMA, confirming that lysine residues were substituted by methacryloyl groups.;This high substitution degree falls within the range reported for GelMA depending on reaction conditions. , Higher GelMA substitution degrees correlate with enhanced mechanical strength, improved tip integrity during skin insertion, and greater structural stability under compression.;In the drug-loaded systems, characteristic peaks of the drugs were observed alongside the backbone spectra of GelMA or PLA. ‖ 结论:Collectively, these findings support the biological plausibility and multifunctional design rationale of the microneedle platform as a candidate strategy for diabetic wound management.
局限
However, these DSC results should be interpreted as supportive evidence rather than definitive proof of improved thermal stability.;These in vitro biocompatibility findings provide strong preliminary evidence supporting further in vivo evaluation of the developed system, particularly in terms of wound closure kinetics, histological tissue remodeling, and angiogenesis-related markers in diabetic wound models.;In summary, all groups exhibited sufficient penetration capacity in this preliminary parafilm model, supporting further evaluation in skin or skin-mimetic substrates, with the A-M-GelMA-MN group achieving the greatest penetration depth and consistency.
