研究问题
Therefore, optimizing cell delivery, enhancing local retention and survival, and boosting the intrinsic reparative potency of stem cells are critical objectives.Nanotechnology has been applied to address these limitations by providing methods to improve stem cell-based therapies [19, 20].
方法
While current mainstay therapies such as phosphodiesterase-5 inhibitors (PDE5i) provide symptomatic relief for many patients, they are often insufficient for those with severe underlying conditions such as diabetes, post-prostatectomy nerve injury, or advanced vascular disease because they do not address the core pathological processes of vascular, neural, and smooth muscle degeneration [3–6].;We examine how nanotechnology is used to improve stem cell delivery and retention, increase therapeutic activity, develop cell-free alternatives based on engineered extracellular vesicles, and create nanostructured scaffolds for tissue engineering.;Labeling stem cells with SPIONs allows them to be visualized as hypointense (dark) signals on MRI, thereby enabling longitudinal monitoring within the same subject.Several studies have applied this technology to ED models.
核心结论
Preclinical studies across various ED models, including cavernous nerve injury (CNI), diabetes mellitus (DM), and aging, have consistently demonstrated significant improvements in erectile function following stem cell administration [8, 9, 11–13].Despite these findings, the clinical translation of stem cell therapy for ED still faces important barriers.;Kim et al. demonstrated that SPION-labeled human MSCs transplanted into the corpus cavernosum of rats with cavernous nerve injury could be detected by MRI for up to four weeks post-injection, with Prussian blue staining confirming the presence of iron-laden cells in the tissue [22].;Wu et al. further demonstrated that this nanotechnology-assisted approach allowed a reduction in the effective cell dose, as lower doses of magnetically targeted ADSCs achieved favorable therapeutic effects, potentially mitigating concerns related to cell sourcing and the tumorigenicity risk associated with high cell numbers [14].Magnetic guidance has also been extended beyond simple cell labeling to biohybrid microrobotic systems.
局限
Therefore, optimizing cell delivery, enhancing local retention and survival, and boosting the intrinsic reparative potency of stem cells are critical objectives.Nanotechnology has been applied to address these limitations by providing methods to improve stem cell-based therapies [19, 20].;However, these imaging findings should be interpreted as evidence of SPION-associated signal rather than definitive proof of long-term viable-cell persistence.;This system combines robotics, nanotechnology, and cell therapy to support spatially controlled delivery.Nanocarrier systems for sustained delivery and improved survivalOnce cells reach the target tissue, survival within ischemic, inflammatory, and oxidative penile tissue becomes another major limitation.
