研究问题
While this study provides a detailed, hypothesis-generating framework of candidate active compounds, targets, and pathways, these network predictions do not constitute direct empirical proof; rigorous in vitro, in vivo, and clinical validation remain necessary to confirm whether these predicted interactions and mechanisms occur in biological systems. From a systems biology perspective, this analysis identifies specific molecular constituents and targets underlying HJD's multi-component and multi-target actions.;Whether amyloid clearance, immune regulation, or autophagy is the primary therapeutic mechanism remains unresolved.;Future studies should clarify whether anti-inflammatory effects produce sustained neuroprotection through interactions with Aβ clearance, synaptic preservation, and neuronal regeneration.3.2.4Bioinformatics study on LDDIn a bioinformatics study, Zhao et al.66 reported that LDD can regulate immune cell infiltration through multi-component, multi-target, and multi-pathway mechanisms, providing a novel perspective for AD immunotherapy.
方法
1Pathogenic mechanism of Alzheimer's disease (AD)1.1Biomedical hypothesisSince beta-amyloid protein (Aβ) was identified as a monomer that forms insoluble fibrils and plaques in the brains of AD patients, research has focused on its neurotoxicity.;Furthermore, accumulating evidence suggests that HJD can modulate gut microbiota dysbiosis and reduce associated Aβ accumulation, attenuate neuroinflammatory responses, and improve cognitive dysfunction.48,492.2Recent research on HJD in AD therapy2.2.1Studies of HJD in animal modelsGu et al.49 demonstrated that HJD suppressed gut microbiota dysbiosis and associated Aβ accumulation, attenuated neuroinflammatory responses, and ameliorated cognitive impairment in APP/PS1 mice.;These limitations substantially constrain confidence in reproducibility and clinical translation despite the promising therapeutic signal.Sun et al.35 reported that HJD modulated microglial activation and improved spatial learning and memory in APP/PS1 mice.
核心结论
Comparison of biomedical and TCM perspectives reveals that both frameworks recognize dementia as a multifactorial, systemic, and progressive disorder.;Furthermore, accumulating evidence suggests that HJD can modulate gut microbiota dysbiosis and reduce associated Aβ accumulation, attenuate neuroinflammatory responses, and improve cognitive dysfunction.48,492.2Recent research on HJD in AD therapy2.2.1Studies of HJD in animal modelsGu et al.49 demonstrated that HJD suppressed gut microbiota dysbiosis and associated Aβ accumulation, attenuated neuroinflammatory responses, and ameliorated cognitive impairment in APP/PS1 mice.;These factors should be considered when interpreting the therapeutic and translational significance of HJD.Fan et al.51 demonstrated that HJD ameliorated cognitive deficits in APP/PS1 mice fed a high-fat diet, likely through broad suppression of inflammation and lipid accumulation.
局限
The principal limitations are small and variable sample sizes, unclear randomization/blinding, reliance on a single behavioral paradigm and AD model, lack of independent replication and causal microbiome validation, and inadequate direct safety assessment.;These limitations substantially constrain confidence in reproducibility and clinical translation despite the promising therapeutic signal.Sun et al.35 reported that HJD modulated microglial activation and improved spatial learning and memory in APP/PS1 mice.;Its main strengths are the integrated multimodal design and formulation characterization, whereas small sample sizes, incompletely reported bias-control procedures, limited safety data, lack of independent replication, and uncertain clinical dose equivalence represent important limitations.Qi et al.50 showed that HJD significantly reversed dysregulated sphingolipid metabolism in both APP/PS1 mouse brains and Aβ-stimulated BV2 microglial cells, while decreasing Aβ deposition and promoting microglial clustering around Aβ plaques.
